PROBLEM TO BE SOLVED: To provide methods and kits for isolating nucleic acid that do not use significant concentrations of flammable components.SOLUTION: A method of eluting RNA from an iron oxide support material includes the steps of: a) bringing a test sample into contact with an iron oxide support material and a binding buffer to form RNA/iron oxide support material complexes, wherein the binding buffer contains a chaotropic agent, less than 15% of an organic solvent, and surfactant, the binding buffer has pH of 3 to 12, and the flash point of the binding buffer is higher than 54.4°C (130 degrees Fahrenheit); b) separating the complexes from the test sample; c) washing the complexes with a solvent-free solution; and d) bringing the complexes into contact with a phosphate-containing buffer. The present invention also provides a method to use the RNA directly in an amplification reaction, without replacing an elution buffer by the method of eluting RNA from the iron oxide support material.SELECTED DRAWING: Figure 1A |
