METHOD FOR OBTAINING PROTEIN GROWTH FACTORS, A GROWTH FACTOR AND FIBROBLAST PROLIFERATION INHIBITOR

The invention relates to medicine. The inventive method for obtaining a protein growth factor consists in producing a conditioned medium and in cultivating fragments, wherein for producing said conditioned medium the fragments of the embryonal bone tissue of a human embryo of a 10-12 gestation weeks are used, the bone fragments are cultivated in a RPMI medium comprising 10 % of embryonal calf serum at a temperature 37 °C, a 100 % air humidity and a 5 % CO₂ content in the atmosphere during 8-10 weeks. Said method also consists in weekly replacing the totality of the medium, in subsequently separating a protein fraction exhibiting a fibroblast-fibrous inhibiting activity by means of known protein cleaning methods, including an exchange chromatography, gel-filtration and centrifuging on a membrane whose pore diameter ranges from 10 to 100 kilodaltons. The thus obtainable protein growth factor is embodied in the form of a purified protein whose molecular weight is equal to approximately 100 kilodaltons, which is water-soluble, thermally stable to a temperature of +56 °C and which exhibits a fibroblast-inhibiting activity, whose degree of manifestation ranges from 40 % to 0 %, and an inhibiting effect when a line raises in a cultural medium containing 30 % conditioned embryonal bone tissue. Said growth factor is used in the form of a fibroblast proliferation inhibitor.
