The application concerns polypeptides comprising an immunoglobulin chain variable domain which binds to Clostridium difficile toxin A, TcdA. These are derived from lama antibodies isolated using a phage display library. Modifications were introduced and their effect on the potency and protease stability was assayed. In some embodiments, dimeric and tetrameric constructs, like bihead and quadrahead constructs, were produced.
