PROBLEM TO BE SOLVED: To produce an enzyme in good productivity by a gene recombination operation while suppressing the dropout of a vector from a host bacterium in culture by culturing microbial transformant having enzyme-producing ability in a medium containing a phytic acid compound. SOLUTION: The pHSPLAMY2 obtained by transducing liquefaction-type alkaliα-amylase gene derived from Bacillus sp-KSM-AP1378 strain into an expression vector plasmid pHSP64 or the like is used as a plasmid, and transduced into Bacillus subtilis ISW1214 by a plasmid-transforming method to provide a microbial transformant having an ability for producing an enzyme such asα-amylase. The transformant is cultured in a medium containing phytic acid compound (e.g. calcium phytate) to provide the objective enzyme in improved productivity without using an antibiotic while preventing the dropout of the vector from the transformant.
