PROBLEM TO BE SOLVED: To provide nucleic acids encoding humanized 9E4 antibodies useful for treatment and diagnosis of a synucleinopathy also known as Lewy body disease.SOLUTION: Provided is a nucleic acid encoding a heavy chain variable region of a specified amino acid sequence and a light chain variable region of a specified amino acid sequence. Further, the nucleic acid encodes a heavy chain constant region of a specified amino acid sequence and a light chain constant region of a specified amino acid sequence with or without C-terminal lysine, where the heavy chain constant region is fused to the heavy chain variable region and the light chain constant region is fused to the light chain variable region. The affinity of a humanized antibody (i.e., Ka) encoded by the nucleic acid that specifically binds to human alpha-synuclein is preferably in a range of 5-fold or 2-fold of that of the mouse antibody 9E4. Preferred humanized antibodies bind to the same epitope and/or compete with the mouse antibody 9E4 for binding to human alpha-synuclein.SELECTED DRAWING: None |
