摘要 |
<p>A recombinant HSV which has the ability to kill and wound tumor cells such as a human glioma specifically in vivo and is useful for the treatment of human gliomas or the like, safe, and easy of clinical application. KeM345, a new recombinant HSV, in which a ?-34.5 gene transcription unit expressed by a Musashi 1 promoter is inserted into the region of the ribonucleotide reductase gene in the genome of an attenuated herpes simplex virus(HSV) by means of a homologous recombination technique is constructed, and purified KeM345 is acquired. Since KeM345 is a recombinant virus itself, it can be not only grown only by transmitting it to cultured cells, but also selectively exhibits an excellent cytotoxic effect (cytolytic ability) on malignant gliomas to induce virus replication comparable to wild-type HSVs.</p> |
