PROBLEM TO BE SOLVED: To observe the internal skeleton of a cell that has not been observed heretofore, such as a red corpuscle or a white corpuscle, by an electron microscope, while holding the shape of a membrane. SOLUTION: After a cell is immersed in a solution, which contains at least one immobilizing agent selected from a group consisting of formaldehyde, paraformaldehyde, trioxane, cacodylic acid and glutaraldehyde, to be immobilized, a surfactant solution and/or an alcohol solution and the cell are mixed/stirred to remove lipid from the cell, and ion etching or ultrasonic treatment is subsequently performed to expose a cell skeleton. Thus, from a viewpoint for efficiently and certainly removing an intercellular substance component from the cell, it is preferable to mix/stirr the alcohol solution and the cell, after the surfactant solution and the cell are mixed/stirred.
