PROBLEM TO BE SOLVED: To provide isolated specific binding members, particularly antibodies, and fragments thereof, to epidermal growth factor receptor (EGFR) in which amino acid sequence is modified or substituted by aberrant post translational modification or aberrant expression, which is found in tumorigenic, hyperproliferative, or abnormal cells but can be generally not detected in normal or wild type cells, and to provide diagnosis and treatment for cancer using the specific binding members.SOLUTION: There is provided an isolated antibody capable of binding to EGFR on tumors having amplification of the EGFR gene, wherein cells of the tumors contain multiple copies of the EGFR gene, and on tumors that express the truncated version of the EGFR receptor de2-7, wherein the antibody does not bind to the de2-7 EGFR junctional peptide consisting of the specific amino acid sequence; wherein the antibody binds to an epitope within the sequence (specific sequence) of residues 287-302 of human wild-type EGFR; and wherein the antibody does not comprise a heavy chain variable region sequence having the specific amino acid sequence and does not comprise a light chain variable region sequence having the specific amino acid sequence.SELECTED DRAWING: None |
