PROBLEM TO BE SOLVED: To provide compositions and methods for achieving sustained therapeutic drug concentrations in a subject.SOLUTION: Provided in the specification are compounds and methods for achieving a sustained therapeutic effect of small molecule anti-cancer agents when administered in vivo. Provided in the specification is a method of achieving a metronomic dosing profile in vivo upon a single administration of a topoisomerase inhibitor prodrug. The method comprises a step of administering a prodrug of a topoisomerase inhibitor to a mammalian subject (such as a human), wherein the topoisomerase inhibitor is releasably attached to a polyethylene glycol moiety, to thereby achieve in a single dose of the prodrug, wherein plasma levels of the topoisomerase inhibitor or a metabolite thereof remain above the level of detection for at least seven days after administration.SELECTED DRAWING: Figure 1 |
