The invention provides a method and device for determining whether variable-region amplification primers have deviation or not and application of the method and device. The method includes the steps of firstly, using a universal primer group to perform multiple PCR amplification on nucleic acid samples containing variable regions to obtain first amplification products; secondly, performing first sequencing on the first amplification products to obtain a first sequencing result; thirdly, using a candidate primer group to perform multiple PCR amplification on the first amplification products to obtain second amplification products; fourthly, performing second sequencing on the second amplification products to obtain a second sequencing result; fifthly, comparing the second sequencing result with the first sequencing result to determine whether the candidate primer group have deviation or not. By the method, deviation of amplification among different primers can be determined effectively, and obtained deviation results are true and reliable. |
