<p>Staining adn sealing a biological specimen which has been applied to a microscope slide and air-dried comprises (a) producing a viscous soln. of a transparent resin in a solvent mixt. of an organic cpd. and water, (b) coating one side of a covr slip with a biological stain, (c) applying viscous soln. to an area of the slide which is smaller than the area of the cover slip, (d) arranging the cover slip on the viscous soln. so that its stain-coated side is in contact with the viscous soln. and (e) leaving the cover plate undisturbed on the soln so that the surface tension can pull the cover slip towards the slide spread the soln. over the specimen and transfer stain from the cover slip to the specimen. The viscous soln. having a viscosity of >500 cp and 3000 cp. Used in preparation of microscope slides of biological (e.g. blood) specimens. Staining an sealing are performed in a single simple step involving only application of the viscous resin soln. to the slide and placing of the cover slip over the applied resin soln. Surface tension pulls the cover slip towards the slide and causes the resin soln. to be spread over the area of slide covered by the cover slip. Dye from the cover slip is transferred through the soln. to the specimen.</p> |
