PURPOSE:To obtain a urokinase useful as a remedy for thrombosis and cardial infarction, in high yield and purity, by adsorbing urokinase to a water-insoluble carrier having a specific group as a ligand, and eluting the adsorbed urokinase in the presence of a nonionic surfactant. CONSTITUTION:Crude urokinase is adsorbed to a water-insoluble carrier having the group of formula (X is OH, CH3, phenyl or amino; n is 3 or 4) (e.g. C4H9, C4H8NH2, etc.) (e.g. agarose, cellulose, etc.), preferably in the presence of a salt, especially a univalent neutral salt (e.g. NaCl, KCl, etc.) to increase the hydrophobic property of the solvent. The adsorbed urokinase is eluted and purified in the presence of a nonionic surfactant. The carrier used in the present adsorption process has improved selectivity of urokinase, and the eluent is effective to elute the urokinase in high efficiency. Accordingly, the process is substantially advantageous as the industrial process for the production of urokinase.
