PURPOSE:To make the exact quantitative determination of a pyruvic acid from triglyceride by consuming the already existing pyruvic acid, then adding ATP or/and chelate agent to a speciment to stop an iCDH reaction, adding enzyme to form the pyruvic acid thereto and measuring the formed pyruvic acid. CONSTITUTION:LDH, NADH, isocitric acid, metallic ions such as magnesium ions or manganese ions and iCDH are added and mixed to and with the specimen to consume pyruvic acid and free glycerol existing already in the mixed soln. The ATP or/and chelate agent are then added thereto to stop the iCDH reaction and the enzyme or enzyme group to form the pyruvic acid as the resulted product of reaction is added to the liquid. The formed pyruvic acid is quantitatively determined. The change from the formula I the formula II is made by the addition of the ATP or/and chelate agent. The reaction of the enzyme group to form the pyruvic acid is exemplified by the formula III. The exact quantitative determination of the material to be examined is thus executed by decomposing the material to be inspected in the specimen and consuming the NADH by the reaction NADH NAD<+>.
