PURPOSE:To assay an objective substance with high sensitivity, by containing either one of apoenzyme and the corresponding coenzyme in a microcapsule receiving lytic action by antigen/antibody reaction and containing the other one outside the microcapsule. CONSTITUTION:Apoenzyme and coenzyme are separately received inside and outside a microcapsule. For example, as the microcapsule, a red corpuscle is used and apoglucose oxidase is used as the substance included in the microcapsule an the absorbancy of the reaction solution is measured at a wavelength of 600nm. 10mul of the microcapsule having anti-T4 antibody preliminarily bonded to the surface thereof and 10mul of a solution containing a complement, coenzyme and glucose are mixed with 10mul of a solution containing T4 at known concn. to perform reaction for 20min. After 20min, 20mul of a solution containing peroxidase and 4-aminoantipyrine diethylaniline and 950mul of a 50mMKH2PO4 buffer solution are mixed with the reaction solution to perform reaction for 20min. By this method, absorbancy increases as T4 increases and a T4<-> calibration curve can be formed.
