PURPOSE: To determine the presence or quantity of IgA1 having O-linked oligosaccharide in a fluid sample by determining the presence or quantity of a label linked or not linked with an anti-IgA/IgA1 /lectin complex. CONSTITUTION: A sample is brought into contact with an anti-IgA antibody to produce an anti-IgA/IgA comp-tex which is then brought into contact with a labeled lectin to produce an anti-IgA/IgA1 /lectin complex. The labeled lectin can be linked with DGa1β(1-3)DGa1NAcIgA1 but can not linked with a sialylated IgA1 or IgA2 lacking O-linked oligosaccharide. Consequently, the presence or quantity of IgA1 immunoglobulin in various humor samples, e.g. serum, subjected to change can be determined by determining the presence or quantity of linked or not linked label and any label, including enzyme, radioactive label, fluorescent molecule, etc., can be employed so long as it can produce a detectable signal.
