PURPOSE:To obtain macro-proteoliposome by a simple process without deactivating and modifying protein by freezing an alkali metal salt solution containing protein and fat and melting the frozen solution to perform dialysis using a salt solution low in osmotic pressure. CONSTITUTION:When an alkali metal salt solution of potassium or rubidium containing protein and lipid is frozen, lipid and protein form a micellar composite in the solution concentrated by freezing to be dissolved therein. When the frozen salt solution is melted at room temp., the protein-lipid composite generates micelle-lamella transformation to become a two-molecule membrane. Further, this salt solution is dialyzed to a salt solution or buffer solution having osmotic pressure lower than that of said solution. Whereupon, large proteoliposome having a diameter of 5-50mum in usual but having a diameter of 300mum according to a condition is obtained. |
