A human superoxide dismutase (Cu,Zn-SOD) is prepd. by (a) destructing cellwall of the recombinant yeast and centrifuging to obtain the Cu, Zn-SOD-contg. crude water-soluble protein soln., (b) adding acetic acid salt into the obtd. soln. and mixing, (c) adjusting the mixed soln. to pH 3.5-5.5 with 3M acetic acid or HCl, (d) centrifugint the obtd. soln. to obtain the supernatant, and (e) passing the obtd. supernatant through S-sepharose column and AcA 54 ultra gel column.
