PURPOSE:To obtain the subject proliferating agent for a vascular endothelial cell, containing interleukins 6 and 8 as an active ingredient, having promoting action on proliferation of the vascular endothelial cell and useful as a curing promoter for burns or wounds, etc., a tissue reparative agent in inflammation and a therapeutic agent, etc., for angiopathy. CONSTITUTION:A human fibroblast is inoculated into a culture medium containing 5% neonatal calf blood serum, bonded to 0.3% microcarrier in a glass culture layer and cultured at 37 deg.C for 5 days while being stirred and the culture medium is exchanged for a serum-free culture medium. Human interferon beta and poly(I):poly(C) are added to continue the culture. The microcarrier is then settled and the supernatant is collected to provide a purified stock solution of interleukins. The resultant stock solution is subjected to column chromatography using silica as a material and heparin as a ligand, reversed phase chromatography and the interleukin 6 is adsorbed on a hydrophobic resin and separated to elute adsorbed substances. The resultant eluate and liquid passed therethrough are respectively subjected to high-performance liquid chromatography to respectively afford interleukins 6 and 8, which are blended with a carrier, excipient, etc., and formulated into a pharmaceutical. Thereby, the objective proliferating agent for the vascular endothelial cell is obtained. |
