NEW MATERIAL:A DNA sequence containing a promoter region of a chromosomic DNA-dependent killer KHS gene derived from Saccharomyces.cere visiae, etc., and a protein coding region. USE:For mass production of chromosome-dependent killer proteins having antimicrobial killer activity. PREPARATION:For example, a genomic DNA is extracted from Saccharomyces.cerevisiae No. 115 strain according to a conventional method and the resultant genomic DNA is then treated with a restriction enzyme and partially hydrolyzed. A nucleic acid fragment of 4-10kbp is separated and collected therefrom by agarose electrophoresis and the obtained DNA fragment is linked to an Escherichia coli-yeast shuttle vector and transduced into Escherichia coli to prepare a genetic bank. A vector is subsequently recovered therefrom to carry out transformation of a host. Screening is then performed in a killer activity detecting culture medium to select a positive strain. A plasmid is subsequently recovered from the aforementioned strain to afford the objective DNA containing the killer KHS gene. |
