Claimed is the use of a DNA vector for producing a medicament for gene therapy where the vector modulates, corrects or activates the expression of an endogenous gene or the expression of a gene introduced by the vector. When intended for treating mammals and humans, the vector lacks an active gene for antibiotic resistance; when intended for treating humans it includes a resistance gene for hygromycin, chloramphenicol, spectinomycin, blasticidin S, phleomycin, bleomycin, puromycin, apramycin or tetronasin. Also claimed are DNA vectors contg. a cystic fibrosis (CF) gene which can be expressed in mammals, or contg. a gene or gene fragment that modulates, activates or corrects an endogenous CF gene and further defined as comprising: (a) an inactive antibiotic resistance gene or an auxotrophic marker; or (b) a resistance gene for one of the antibiotics listed above. |
