<p>PROBLEM TO BE SOLVED: To obtain the subject new monoclonal antmbody recognizing 14-3-3 protein as an antigen, specifically reactive with human pulmonary cancer tissue and human pulmonary cancer cell in sputum and useful as a reagent for the cytological examination of human pulmonary cancer. SOLUTION: This antibody is a new mouse monoclonal antibody recognizing 14-3-3 protein as an antigen, reactive with human pulmonary cancer tissue and human pulmonary cancer cell in sputum, belonging to immunoglobulin class IgG and usable e.g. for the diagnosis of human pulmonary cancer as a reagent for the cytological examination of human pulmonary cancer. The monoclonal antibody can be produced by separating purified 14-3-3 protein from bovine brain by reverse-phase high-performance liquid chromatography, injecting the protein together with a complete Freund's adjuvant into the abdominal cavity of a BALB/c mouse to effect the immunization of the mouse, collecting the spleen cell after the final immunization, fusing with a myeloma cell, subjecting the fused cell to selective culture in HAT medium, cloning the obtained hybridoma and culturing the product.</p> |
