PROBLEM TO BE SOLVED: To obtain the subject new plasma membrane-located sialidase having a specific amino acid sequence, having an activity for removing a sialic acid residue from the non-reducing terminal of genglioside, and useful as a sugar chain research reagent, a genetic treatment medicine, etc. SOLUTION: This new plasma membrane-located sialidase protein comprises a protein having an amino acid sequence of formula I or formula II, or a protein which has the amino acid sequence of formula I or formula II wherein one or several amino acid are substituted, deleted, inserted or transferred and which has an activity for removing a sialic acid residue from the non-reducing terminal of ganglioside. The protein is useful as a sugar chain research reagent, a genetic treatment medicine, etc. The protein is obtained by homogenizing a bovine brain in a buffer solution, centrifuging the homogenized solution, collecting the supernatant, further centrifuging the supernatant, suspending the precipitate fraction in a buffer solution, adding 5% deoxycholid acid, sufficiently homogenizing the mixture, and subsequently purifying the supernatant by an affinity chromatography method. |
