摘要 PROBLEM TO BE SOLVED: To provide a method for detecting a nucleic acid and a target sequence, especially the method for detection using a fluorescent quenching. SOLUTION: A nucleic acid for detection is used for detecting a target sequence of a nucleic acid with a fluorescent quenching system. The nucleic acid for detection contains at least two oligonucleotides and is partially a single strand and partially a double strand. In a donor/acceptor dye pair, either one of the two dyes of the donor/acceptor dye pair is bound to the first oligonucleotide and the other is bound to the second oligonucleotide. When the first and the second oligonucleotides are base paired, both approach to quench the donor fluorescence. When the target is absent, a part of the detected oligonucleotide retains the form of the single bond and a part thereof retains the form of the double strand. When the target is present, the nucleic acid for the detection is replaced from the first nucleotide and the distance between the donor and the acceptor dyes is increased to cause a change in fluorescence.
