<p>PROBLEM TO BE SOLVED: To carry out stable mass proliferation of the tissue of an orchidaceous plant by excising a tissue containing a growth point of a shoot obtained by formation thereof according to a specific method and carrying out the tissue culture. SOLUTION: An inflorescence of an orchidaceous plant is used as an explant body and the tissue culture of the resultant explant body is carried out to form each shoot. A tissue containing the growth point of the obtained shoot is then excised and subjected to the tissue culture. In the process, a method for the tissue culture after excising the tissue containing the growth point can be performed by conducting the tissue culture in a solid culture medium containing 1-naphthaleneacetic acid(NAA) and 6-dibenzyladenine (BA) or/and 1- phenyl-3-(1,2,3-thiadiazol-5-yl)urea (TDZ) as phytohormones and then carrying out the tissue culture in a liquid culture medium containing the NAA and BA or/and TDZ or can be conducted by performing the tissue culture in a solid culture medium containing the NAA and BA or/and TDZ in a 1/2 Murashige- Skoog culture medium, subsequently forming the shoot in the tissue and continuously carrying out the tissue culture of the resultant tissue in the solid culture medium.</p> |
