PROBLEM TO BE SOLVED: To readily discriminate the sex of fish such as Paralichthys olivaceus in a step of a fry in fish farming by preparing a biological sample of the fish, and detecting an expression pattern of a mRNA of a gene having the sex difference in the expression in the biological sample. SOLUTION: A biological sample comprising gonad of fish (e.g. Paralichthys olivaceus), the whole body of the fish, or the like is prepared, and an RNA is extracted from the biological sample. A cDNA is synthesized from the obtained RNA. An amplifying reaction of the cDNA is carried out by using a primer comprising an oligonucleotide containing at least a part of a sequence complementary to a base sequence of an aromatase gene which is the gene having sex difference in expression, and the expression pattern of the mDNA of the aromatase gene is detected to discriminate the sex of the fish readily at the early time to detect the ratio of the sexes of released seeds at the step of fries before the release.
