PURPOSE: Methods for large scale purification of neurotrophins, including mature NGF, suitable for clinical use provide means to separate neurotrophins from various less desirable misprocessed, misfolded, size, glycosylated, or charge forms. CONSTITUTION: A process to isolate a neurotrophin from a mixture containing other proteins, comprises separating the neurotrophin from the other proteins in the mixture by using hydrophobic interaction chromatography resin, wherein the separating comprises loading the mixture containing the neurotrophin and a misfolded variant, an incorrectly proteolytically processed variant, or a glycosylation variant of that neurotrophin onto a hydrophobic interaction chromatography resin and eluting the neurotrophin from the resin under conditions in which the neurotrophin separates from the variant or variants.
