PURPOSE: Assayable hepatitis C viral replicon, an assayable replicon-containing cell line, and a screening method of HCV inhibitor using the same are provided. Therefore, the accuracy of HCV replication detection is improved by using assayable hepatitis C viral replicon which effectively replicates within higher cells. CONSTITUTION: Assayable hepatitis C viral replicon(KCTC 10309BP) comprises internal ribosomal entry site(IRES), a selection marker coding sequence, IRES of encephalomyocarditis(EMCV), and an expressably linked reporter protein encoding sequence at the 5'-terminal, wherein the reporter protein is selected from renilla luciferase, firefly luciferase, green fluorescence protein, and secreted alkaline phosphatase. A transformed liver cell line(KCTC 10310BP) with RNA transcriptome produced from the DNA replicon of HCV contains the assayable hepatitis C viral replicon(KCTC 10309BP). A screening method of HCV inhibitor comprises the steps of: (a) treating the assayable replicon-containing cell line with a replication inhibitor and culturing the cell line; and (b) measuring the expression level of the reporter protein.
