<P>PROBLEM TO BE SOLVED: To provide a method for eluting a protein, by which the protein, such as an enzyme, having functions liable to be affected by pH environments can be recovered without causing the deterioration in the functions of the enzyme, while controlling and retaining the pH environment on the recovery within the pH-stable region of the enzyme. <P>SOLUTION: This method for eluting the protein by an isoelectric focusing method comprises the following processes. (a) A process for forming a hole at a prescribed position of a gel which is used for the isoelectric focusing and at whose both ends a cathode and an anode are disposed, (b) a process for charging an adsorbent for catching the protein into the hole, (c) a process for bringing a specimen containing the protein to be eluted into contact with the inside of the adsorbent or with the adsorbent, (d) a process for applying an electric field to the gel to adjust the pH values of the adsorbent to pH values corresponding to a gel pH gradient corresponding to the position of the hole, thereby eluting the protein contained in the specimen and adsorbing the protein to the adsorbent, and (e) a process for recovering the protein from the adsorbent. <P>COPYRIGHT: (C)2004,JPO |
