METHODS FOR MAKING AND BREAKING PRIONS (PRPSC) PRODUCED FROMNATIVE PRIONS (PRPC), IN VITRO AND FROM RECOMBINANT PRION PROTEINS

Agents for making and breaking prions (PrP Sc) under "physiological" conditions, in vitro, are revealed. These include a synthetic copy of a domain of a bovine/ovine naturally occurring protein ("cf22aa") that converts constitutive native hum an brain prion proteins (PrPC) to (PrP Sc), the other, a synthetic copy of a domain of a protein encoded on the anti-sense strand of human P53 gene, ("pbf13aa") breaks-up PrP from T SE infected human brains, and PrP Sc artificially added to human blood and othe r fluids. Also revealed are two mono-specific antibodies which block conversion, to Pr P Sc and remove excess of pbf13aa from fluids treated with this polypeptide, respectively.
