Human serum albumin particles which are suitable for binding technetium are produced. Human serum albumin purified by dialysis is adjusted to a pH of 9.8 to 10.2 and kept at 71 to 73 DEG C for 10 to 20 minutes. It is then cooled to 45 to 50 DEG C, adjusted to a pH of 6.8 to 7.0 with acid and 0.17 to 0.22% by weight of tin(II) ions, based on the albumin, is added. The pH is adjusted to 5.2 to 4.8 by further addition of acid and a further 1.53 to 1.73% by weight of tin(II) ions, based on the albumin, are added. The temperature is raised to 71 to 73 DEG C and the suspension is subjected to countercurrent sedimentation with physiological saline solution. After decantation of the saline solution, the concentration of the particles is adjusted to 2.5 to 3.0 mg of albumin per millilitre of suspension with sterile distilled water. The suspension is then freeze-dried in vessels precooled to -80 DEG C. The freeze-dried human serum albumin particles can be labelled immediately before use with technetium-99m by adding isotonic sterile pertechnetate solution and shaking briefly. |
