<p>In the prodn. of liposomes which are cytotropic and have improved permeability for the administration of medicaments due to having enzymes (e.g. neraminidase) and cell specific immunoglobulins and/or other cell-specific factor coupled covalently or adsorptively to the liposome membrane, turbo-mixers with stator and rotor are used in the liposome prodn. esp. in the encapsulation of organ and cell extracts, opt. after elimination of the unincorporated soln. by filtration, dialysis or centrifugation and before coupling of the cytotropic substances to the membrane. The liposomes are suspended in aq. solns. of proteins, proteids, peptides or macromolecular organ extracts or antibody sera with or without the addition of detergents. The liposome formulations can be used e.g. for inhibiting specific-cell functions (e.g. secretion of insulin or other hormones) by incorporating appropriate inducers or inhibitors in the liposomes and coating the liposome membrane with cytotropic substances to limit the action of the inhibitor or inducer to the target cell. The formulations can also be used in cancer therapy use of a turbo-mixer simplifies prodn. of the cytotropic liposomes. An example describes the prodn. of egg lecithin/cholesterol/diethyl phosphate liposomes using an Ultr-Turrax (RTM) turbo-mixer.</p> |
