3-Hydroxy-ML-236B of formula (I) known as M-4 and its salts, esters and cyclic lactones were prepd. by reaction o the ML-236B deriv. with a hydroxylation enzyme, which produced by Nocardia spp. Thus, N. autotrophica subsp. ametistina FERM P-6183 was inoculated into a medium contg. glucose 1.0, peptone 0.2, meat ext. 0.1, yeast ext. 0.1, corn steep liquor 0.3% and tap water. Na ML-236B carboxylate was added to a total of 0.05% w/v and fermn. was continued at 26↿C for 2 days with agitation, and continued another 5 days to give culture broth contg. M-4 and M-4'; deriv. (I) inhibit the biosynthesis of cholesterol, and have comparable activity to that of ML-236B, and are used for the treatment of hyperlipaemia.
