PROBLEM TO BE SOLVED: To produce an alkaline phosphatase, capable of manifesting high catalyst activities and useful as a labeling enzyme, etc., in an enzyme immunoassay. SOLUTION: This alkaline phosphatase (EC 3.1.3.1) is obtained by culturing a Vibrio sp. SMY-2 strain (FERM P-15019), etc., and has the following physicochemical properties: (1) capable of catalyzing a reaction of a phosphoric monoester + H2 O... an alcohol + inorganic phosphoric acid; (2) >=6000/sec rate constant of the catalytic reaction based on the subunit at 37 deg.C when p- nitrophenyl phosphate is used as a substrate; (3) optimum pH: about 10.5 (at 37 deg.C) and stable at pH7-8; (4) optimum action temperature: 30-45 deg.C [in a diethanolamine (DEA) buffer solution at pH10.0] and (5) molecular weight: about 58000 [measured by a sodium dodecyl sulfate-polyacrylamide gel electrophoresis(SDS-PAGE)]. |
