PROBLEM TO BE SOLVED: To determine the activity of the subject enzyme which has an important role in biosynthesis of saccharides and is useful as a tumor marker by using a fluorescent saccharide derivative which can be an acceptor of sialyltransferase or the like as a substrate. SOLUTION: A saccharide derivative represented by the formula: R1 -O-X-Y (R1 has a saccharide chain having 2-10 saccharide residues and a structure that can be an acceptor of sialyltransferase or fucosyltransferase; X is a 1-4C alkylene; Y is a fluorescent substituted amino; O atom bonds to the 1-carbon atom in the reduced terminal saccharide residue), typically 2-pyridylaminoethylβ-lactoside, is used as a substrate and is allowed to react in the presence of a sugar nucleotide as a donor with an action of sialyltransferase or fucosyltransferase. After the reaction is stopped, the reaction product is separated and determined by HPLC to measure the activity of the objective glycosyltransferase.
