1356413 Purified Australian antigen PFIZER Ltd 7 Sept 1972 [9 Sept 1971] 42017/71 Heading A5B Purified Australian antigen is obtained by an isopyknic banding technique whereby a blood serum containing the antigen is contacted with the high density end of a liquid medium having a density gradient therein which includes the density of the antigen (c. 1.2), and subjecting the medium and serum to ultracentrifugation until an equilibrium distribution of serum components is obtained and thereafter separating the fraction rich in Australian antigen. Suitable liquid media are Cs Cl and KCl aqueous solutions. Further purification may be achieved by dialysis and further ultracentrifugation in contact with another liquid medium having a density gradient (e.g. sucrose solution) until equilibrium is not achieved; but the serum components, including the antigen, are distributed throughout the medium, the required fractions being subsequently collected.
