In examples, the title ketone is reacted with NH 2 OH to form its oxime (III) or with isoamyl nitrite/HCl to form (IX); (III) gives an O-ptoluenesulphonate which is reacted with NaOEt to give (V). (V) is also prepared by reducing (IX) with H 2 /Pd-C. (VII), prepared by reacting (V) with an appropriate haloalkanoyl acid chloride, react with NHR1R2 giving (I). Therapeutic compositions having antitussive activity comprise (I) and are preferably administered orally, although the parenteral and rectal routes are also mentioned.
