PURPOSE:To prepare uniform liposome efficiently and in large amt. by stirring liposome membrane composing substance with aq. soln. at a temp. above the phase transfer temp. of the membrane composing substance itself. CONSTITUTION:About 10-1,000pts.wt. aq. soln. such as water is added to 1pt.wt. liposome membrane composing substance such as phosphatidyl choline, etc., and the mixture is stirred at a temp. above the phase transfer temp. of the membrane composing substance itself. If desired medical substance (e.g. penicillin G) is previously added to said aq. soln. in this stage, uniform liposome incorporated with the desired medical substance is prepd. by the above described stirring with good reproducibility and in large amt. Since no org. solvent is used in this method for prepg. liposome, there is no problem in public peace, operational safety, and biological safety. Moreover, the prepn. is performed quite easily requiring only temp. control during preparation. Further, it is possible to prepare liposome preparation having high content of medical substance. Expansion of preparation scale is easy, therefore, commercial production of liposome preparation is possible. |
