PURPOSE:To produce a plasminogen activator, by propagating a normal diploid fibroblast in a culture medium and cultivating the fibroblast in a serum-free medium with a producing inducer such as phospholipase A2 and peptones in high cell density added. CONSTITUTION:A normal diploid fibroblast is cultivated and propagated in a serum medium or serum-free medium. Then the fibroblast is cultivated in a serum-free medium added by at least one producing inducer selected from phospholipase A2, phospholipase C, fradykinin, concanavalin A and phytohemagglutinin and peptones in high cell density. Thus a plasminogen activator having 65,000-72,000 molecular weight according to measurement by SDS-polyacrylamide gel electrophoresis as well as characteristics as a serine protease suffering activity inhibition by p-aminobenzamidine, etc., according to classification as a protease can be produced. |
