<p>Prodn. of fructose from inulin is effected by hydrolysis with microbial inulase (I) in an aq. medium at 60-65 deg.C and pH 3.5-7, where (I) has an endo-inulase/exo-inulase wt. ratio (R) of 1:10 to 10:1 (pref. 1:7 to 7:1), esp.1:2 to 4:1 and is used in an amt. sufficient to obtain complete hydrolysis of the inulin. (I) is pref. obtained by culturing a suitable strain of Aspergillus ficum, esp. CBS 55565, Hydrolysis is pref. effected at 60 deg.C and pH 4.5-5.0 for 2-4 days using 0.25-10 (esp. 1-5) SU of (I) per g of inulin. (I) with the indicated R values has better thermal stability than prior art inulase prepns. and can thus be used at higher temps., thereby facilitating dissolution of the inulin, improving compatibility with conventional saccharification processes, and reducing the risk of microbial infection.</p> |
