PURPOSE: To simply and rapidly obtain a sample for analyzing a saccharide chain such as high-accuracy diagnosis, etc., for rheumatism by adding a proteolytic enzyme and a saccharide chain liberating enzyme simultaneously or without placing an interval, excising the saccharide chain, labeling the saccharide chain and then purifying the labeled saccharide chain. CONSTITUTION: Three of a proteolytic enzyme such as a papain, a trypsin or a chymotrypsin, a saccharide chain liberating enzyme such as a peptide- N<4> -(N-acetyl-β-glucosaminyl)asparagine amidase and a sialidase, etc., are added to a glycoprotein such as an immunoglobulin G, a fibrinogen or an ovalbumin simultaneously or without placing an interval to excise a saccharide chain without containing the sialic acid from the glycoprotein. The resultant saccharide chain is then labeled and subsequently purified with an amino column to simply and rapidly afford a sample for analyzing the saccharide chain such as diagnosis, etc., of diseases such as rheumatism with a high accuracy. Furthermore, the sample is analyzed by high-performance liquid chromatography using an ODS-silica column and the saccharide chain composition of the sample is determined to diagnose the diseases by a change thereof.
