Detecting the presence of an immunologically reactive molecule (IRM) in a sample, comprises: (a) placing the sample into contact with a standard solution ((SS) of one or more immunologically reactive reference molecules (IRRMs)) in an incubation mixture, to enable formation of immune complexes (ICs) between the IRRM and the IRM, and (b) determining whether ICs have been formed by comparing one or more of the physical parameters of the components of the incubation mixture with the same physical parameters of the components of the SS. The physical parameters are molecular weight, charge and form. Step (b) is carried out by: (i) column chromatographic separation of the components of the incubation mixture, (ii) column chromatographic separation of the components of the SS in the same manner, and (iii) comparison of the elution patterns of both separations. A change in the elution speed of the IRRM in the incubation mixture, relative to the elution speed of the same IRRM in the SS, indicates formation of an IC.
