Reagent (A) for determining (i) the number of leucocytes and of polynuclear basophils and (ii) the hemoglobin (Hb) content in a blood sample comprises: (1) a buffer to provide acidic pH, preferably below 3; (2) at least one cationic detergent (I); and (3) a nitrogenous compound (II).
