PROBLEM TO BE SOLVED: To provide a simple, accurate and effective technique for detecting an objective oligonucleotide sequence. SOLUTION: This method detects the existence of the objective oligonucleotide sequence in a nucleic acid present in a nucleic acid sample. The method comprises a process in which the nucleic acid sample is allowed to bond to a polynucleotide reagent under hybridizing conditions. In the process, the sample or any one of the components of the reagent bonds to a supporting body, and the material to be analyzed and the polynucleotide reagent are hybridized to form a label which bonds to the supporting body via a selectable cleavage part: R1 OXOR2 in which X is a periodic-acid cleavable bond.
