NOVEL PLASMID, PRODUCTION THEREOF AND NOVEL BACTERIUM CONTAINING SAME

摘要

PURPOSE:To produce a transformed bacterium, by forming a plasmid integrated with a deoxyribonucleic acid fragment carrying a genetic information participating in extracellular production of CGTase of a bacterium belonging to the genus Bacillus at a restriction site. CONSTITUTION:A DNA fragment providing extracellular production ability of cyclomaltodextrin glucanotransferase (CGTase for short) is prepared by using a restriction enzyme. Vector DNA of plasmid is restricted by the use of a restriction enzyme which does not inhibit genetic information participating in extracellular production of CGTase of the prepared DNA fragment. The restriction site of the restricted vector DNA of plasmid is recombined with the above-mentioned DNA fragment and the recombinant plasmid is extracted. DNA of plasmid pBR322 is preferably used as the vector DNA of plasmid.
