Preparation of human monoclonal antibodies

The preparation of human monoclonal antibodies (MAb) comprises: (a) transplanting a human stem cell (HSC) from a donor into a first animal embryo (E1); (b) transplanting HSC and an animal stem cell (ASC) in a second animal embryo (E2); (c) injecting into a first born tolerant animal (A1) of E1 an antigen (Ag) to stimulate the immune system; (d) isolating human antibody-producing B-cell from A1; (e) fusing of the B-cells obtained from A1 with tumour cells, especially myeloma cells from an animal (A2) derived from E2, to produce hybridoma cells; (f) injecting the hybridoma cells into a twice tolerant A2, and (g) isolating MAb from the resulting ascites, the resulting blood serum, the produced milk or the laid eggs of A2. Also claimed is a method for the production of mouse MAb comprising: (a) transplanting stem cells (MSC) from a mouse (M1) into an animal embryo (E); (b) injecting hybridoma cells from M1 into an animal (A3) derived from E which is mouse-tolerant, and (c) isolating mouse MAb from the ascites, blood serum, milk or eggs from A3.
