A suitable buffer, e.g. to achieve a pH in the range of 5 to 9 is a mixture of dibasic sodium phosphate and monobasic potassium phosphate, The composition may be prepared by adding a buffer to the formalin and thymol and adding to the resulting mixture a freshly prepared isotonic solution of lithium chloride. The treatment of fresh sputum is exemplified.
