摘要 <p>1475608 Automatic analysis using suspended reagent particles TECHNICON INSTRUMENTS CORP 5 June 1974 [8 June 1973] 24996/74 Heading G1B A constituent of a sample is determined by suspending in a liquid a plurality of insoluble particles each particle comprising a reagent, mixing the liquid with the sample to react the sample and the reagent and determining quantitatively a product of the reaction on each particle successively as a measure of the amount of constituent present. The particles may consist of reagent alone, for example enzyme cross-linked with bifunctional compound, or reagent bonded to a support material by covalent of ionic bonds or embedded in a gel matrix support. Support materials may be dextran, acrylic polymer, polyamino acids, polystyrene, starch, cellulose, glass. In the embodiment shown, reagent in stirred flask 18 comprises a suspension in a solution of for example a chromogen insoluble particles of three different particle sizes, each size particle comprising a reagent or reagents for a reaction with a constituent of the samples flowing in line 10, the mixed sample-reagent stream being segmented by air introduced through line 14, incubated at 26, mixed with diluent introduced by line 28 and degassed at junction 34, the liquid stream flowing through absorption detector-light scattering particle size detector assembly 40 which includes sheath stream flow cell 41 for determination of the amount of reaction product on each particle and to count the number of particles of each size passing through the flow cell. The detector assembly includes dichroic mirror 56 which acts as a beam splitter and directs part of the beam through a centrally apertured field stop 58 to a photo-diode 60 which provides a quantitative indication of the amount of, e.g. dark coloured, reaction product on each particle and part of the beam through a reticule 62 forming a dark field stop to a photo-diode 64 which measures scattered light to enable recognition of individual particles according to their size. The signals from photo-diode 64 are amplified and fed to the inputs of four comparators I to IV, Fig. 3 (not shown), each having a different characteristic input reference voltage. The amplified signal indicates a particular particle size when the voltage produced falls between a particular pair of reference voltages. The particles of each size are counted and the total amounts of reaction product carried by a specific number of particles of each size is determined, the results being passed to a printer.</p>
