The invention relates to a process for the determination of an immunologically active material in a sample. The process is characterised in that the sample is brought together with a receptor which is able to bind the immunologically active material specifically, with the immunologically active material, which is labelled with a substance which is able to modify the degree and the nature of the activity of the enzyme, and with an enzyme and an enzyme substrate, whereupon the degree or the nature of the enzyme activity obtained is measured and compared with that obtained using a standard. The invention further relates to a reagent for carrying out the process. A suitable substance which is able to modify the degree or the nature of the activity of an enzyme is in particular methotrexate.
