Automatic column chromatography method for enriching, prepurifying and concentrating substances in heterogeneous liquid substance mixtures

In this method, a substance to be analysed in an extremely heterogeneous mixture of substances is chromatographed by column chromatography. This is done by introducing the mixture of substances in 'forward flow' into a column and, after elution of interfering substances, is eluted in the forward direction. A downstream mixing section makes it possible to alter the fraction containing the substance to be analysed or the substance to be analysed itself so that the substance to be analysed is retained in the second column. After further elution of interfering substances, the fraction containing the substance to be analysed, which has been retained in a narrow zone, is eluted in 'backward flow' from the second column and fed to a downstream chromatographic system or detection system. An electronic circuit reduces the manual effort substantially and results in a high reproducibility.
