PURPOSE:The titled plasmid, having a specific limited cleavage sites for several kinds of restriction enzymes and a specific molecular weight, and suitable for vector plasmid of coryneform bacteria producing glutamic acid. CONSTITUTION:A plasmid pAG3 having about 4.8 kilobase molecular weight and one cleavage site respectively for restriction enzymes HindIII, Pst I, BamH I and Xba I. The plasmid pAG3 can be obtained from Corynebacterium melassecola 22220 (FERM-P No.559). A strain can be improved by genetic manipulation of coryneform bacteria producing glutamic acid by utilizing a vector plasmid prepared by using the plasmid pAG3 or modifying the plasmid pAG3. |
